植物几丁质酶通过降解真菌细胞壁起到抵抗真菌病害的作用,是一种重要的病程相关蛋白。现以苹果品种‘富士’、‘秦冠’及其杂交后代为试材,采用荧光定量PCR的方法,研究接种苹果褐斑病对苹果几丁质酶基因MdChi表达的影响。结果表明:褐斑病菌诱导下MdChi在3份种质的叶片中均有表达,感病品种‘富士’叶片中MdChi基因表达量明显高于抗病品种‘秦冠’及二者杂交后代‘FQ-2’。侵染后24h时‘富士’叶片中的MdChi表达量明显上升,48h后表达量又迅速下降。而抗病品种‘秦冠’及抗病杂种‘FQ-2’中MdChi基因表达量在侵染后的96h内未有较大幅度变化,均保持在较低水平。
Plant chitinase acts as an anti-fungal disease by degrading fungal cell walls and is an important disease-related protein. The apple cultivars ’Fuji’, ’Qin Guan’ and their hybrid progenies were used as materials to study the effect of apple brown spot inoculation on the expression of MdChi in apple chickens by fluorescence quantitative PCR. The results showed that MdChi was expressed in leaves of three germplasms induced by brown spot pathogen. The expression level of MdChi gene in susceptible cultivar ’Fuji’ was significantly higher than that of resistant variety ’Qin Guan’ and their hybrid progenies’ FQ -2’. The expression of MdChi in ’Fuji’ leaves increased significantly at 24h after infection, and the expression decreased rapidly after 48h. However, the MdChi gene expression in the resistant variety ’Qin Guan’ and resistant hybrid ’FQ-2’ did not change significantly within 96 h after infection, and remained at a low level.
