背景:细胞周期素D1和P16通过调节周期素依赖性激酶4的活性而实现促进细胞增殖和抑制细胞分裂的作用。人类多种肿瘤中都存在不等的细胞周期素D1基因扩增或过表达,并伴随P16基因的高频率失活。目的:探讨细胞周期素D1基因的扩增和P16基因的缺失与肺癌的关系。设计:病理标本对照实验。单位:解放军第八一医院生化科。对象:肺癌标本40例均来源于1998-06/1999-06解放军第八一医院手术的肺癌患者,患者均知情同意,分别在肿瘤区及正常肺组织区取材。方法:采用多重聚合酶链反应方法,以提取的基因组DNA为模板,在同一反应管中同时扩增细胞周期素D1基因和P16基因。取10μL聚合酶链反应产物,在15g/L琼脂糖凝胶上电泳分析。主要观察指标:①肺癌患者病理组织中细胞周期素D1和P16的基因扩增结果。②细胞周期素D1和P16与肺癌生物学特性的关系。结果:①细胞周期素D1和P16扩增结果:肺癌细胞周期素D1扩增的阳性率为55%(22/40),P16阳性率为65%(26/40)。②细胞周期素D1和P16与肺癌生物学特性的关系:两者扩增的阳性率与淋巴结转移和临床病理分期密切相关,随转移的发生和病程的发展,细胞周期素D1扩增阳性率升高而P16阳性率下降。结论:细胞周期素D1的扩增和P16的缺失与肺癌细胞的高增殖活性密切相关。检测两种基因的变化,可有助于判断肺癌的转移、恶性度及评估预后。
BACKGROUND: Cyclin D1 and P16 function to promote cell proliferation and inhibit cell division by regulating the activity of cyclin-dependent kinase 4. A variety of human tumors exist unequal cyclin D1 gene amplification or overexpression, accompanied by high frequency of P16 gene inactivation. Objective: To investigate the relationship between cyclin D1 gene amplification and P16 gene deletion and lung cancer. Design: pathology specimen control experiment. Unit: People’s Liberation Army eighth hospital biochemistry. PARTICIPANTS: Forty lung cancer samples were all from lung cancer patients undergoing surgery in the 81st Hospital of People’s Liberation Army from June 1998 to June 1999. All patients were informed consent and were taken in the tumor area and the normal lung tissue area respectively. Methods: Multiplex polymerase chain reaction (PCR) was used to amplify the cyclin D1 gene and P16 gene in the same reaction tube with the extracted genomic DNA as a template. Take 10 μL of polymerase chain reaction products, electrophoresis analysis on 15g / L agarose gel. MAIN OUTCOME MEASURES: ① Gene amplification results of cyclin D1 and P16 in pathological tissues of lung cancer patients. ② the relationship between cyclin D1 and P16 and biological characteristics of lung cancer. RESULTS: ① The results of cyclin D1 and P16 amplification showed that the positive rate of cyclin D1 amplification in lung cancer was 55% (22/40) and the positive rate of P16 was 65% (26/40). ② The relationship between cyclin D1 and P16 and the biological characteristics of lung cancer: The positive rate of amplification between the two is closely related to lymph node metastasis and clinical stage. With the development of metastasis and the course of disease, the positive rate of cyclin D1 amplification High and P16 positive rate decreased. Conclusion: The proliferation of cyclin D1 and the deletion of P16 are closely related to the high proliferative activity of lung cancer cells. Detecting changes in both genes can help determine lung cancer metastasis, malignancy and prognosis.
