目的运用生物信息学方法预测分析弓形虫Toxofilin基因编码蛋白的功能域及三级结构。方法 PCR扩增Toxofilin全长基因序列,利用SignaIP和TMHMM在线网站预测分析其编码蛋白的信号肽、跨膜区;利用DNAStar软件分析Toxofilin蛋白的亲水性、抗原指数;联合运用ExPASY和PRODATA分析Toxofilin蛋白的功能域及三级结构。结果 PCR扩增Toxofilin基因大小为738bp。DNAstar分析其编码蛋白Toxofilin序列存在典型信号肽,但不存在跨膜区,在其N端存在较高的亲水性基团和抗原指数。ExPASY分析Toxofilin蛋白存在酰基化、磷酸化蛋白激酶位点。结论 Toxofilin蛋白含有信号肽和磷酸化位点等功能域,抗原指数,为验证Toxofilin功能奠定了理论基础。
Objective To predict and analyze the functional domains and tertiary structure of the Toxofilin gene encoded by Toxoplasma gondii using bioinformatics methods. Methods The full-length Toxofilin gene was amplified by PCR. The signal peptide and transmembrane region of Toxofilin protein were predicted by using SignaIP and TMHMM online websites. The hydrophilicity and antigen index of Toxofilin protein were analyzed by DNAStar software. The combination of ExPASY and PRODATA Functional domain and tertiary structure of protein. Results The Toxofilin gene was 738 bp in size. DNAstar analysis of its coding protein Toxofilin sequence typical signal peptide, but no transmembrane region exists at its N-terminal higher hydrophilic groups and antigen index. ExPASY analysis of Toxofilin protein presence of acylation, phosphorylation of protein kinase sites. Conclusion Toxofilin protein contains functional domains such as signal peptide and phosphorylation site, antigen index, which laid the theoretical foundation for the verification of Toxofilin function.
