目的设计一种可提高组织切片苏木精-伊红(HE)染色速率及染色质量的病理组织切片染色装置。方法设计制作由水浴箱、带盖染色缸、电控箱三部分构成的组织切片恒温染色台,实验比对组织石蜡切片上的石蜡膜溶于二甲苯的互溶速率和镜下染色效果。结果在37.0℃条件下,石蜡膜在二甲苯中的溶脱时间比室温条件下缩短了近一半的时间,互溶速率两组比较差异有显著统计学意义(P<0.001);在减少1/2染色时间染制的切片组织色泽与室温条件下的染色效果类似,而且细小颗粒着色清晰。结论应用该设备条件的组织切片染色效果优于传统常温方法。
Objective To design a pathological tissue section dyeing device that can improve the tissue section hematoxylin-eosin (HE) staining rate and the dyeing quality. Methods Tissue section thermostat dyeing station consisted of water bath, covered dyeing cylinder and electric control box was designed and manufactured. The mutual solubility rate and microscopic staining effect of paraffin film dissolved in xylene on tissue paraffin section were compared. Results At 37.0 ℃, the elution time of paraffin film in xylene was shortened by almost half compared with that at room temperature. The miscibility rate of paraffin film was significantly different between the two groups (P <0.001) The time-stained tissue sections are similar in color to the staining results at room temperature, and the fine particles are clearly colored. Conclusion The staining results of tissue sections using this equipment are better than those of the traditional room temperature method.
