目的研究人贲门腺癌中Musashi-1、细胞转录因子4(Tcf4)、尾侧型同源转录因子2(Cdx2)的表达及临床意义。方法采用免疫组化法检测贲门腺癌、癌旁组织、正常贲门组织中Musashi-1、Tcf4、Cdx2的表达情况,采用Western blot检测贲门腺癌及癌旁组织中Musashi-1、Tcf4、Cdx2的表达,并分析其表达与临床病理特征、危险因素的关系。结果在正常贲门上皮、癌旁组织、贲门腺癌中,Musashi-1阳性表达率分别为20.0%、45.0%、79.3%(P<0.05),Tcf4的阳性表达率分别为20.0%、40.0%、75.9%(P<0.05),Cdx2的阳性表达分别为20.0%、60.0%、37.9%(P>0.05);Western blot结果显示Musashi-1、Tcf4在癌组织中的表达高于癌旁组织,Cdx2在癌旁组织中的表达高于癌组织;Musashi-1与Tcf4在贲门腺癌组织中呈正相关(r=0.662,P<0.05),Tcf4与Cdx2在癌旁组织中呈弱相关(r=0.375,P<0.05),Musashi-1与Cdx2在癌组织中无明显相关(P>0.05);Musahi-1、Tcf4、Cdx2的表达主要与肿瘤组织分化程度有关(P<0.05);Musahi-1、Tcf4、Cdx2的表达与吸烟、幽门螺杆菌感染、胃食管反流病密切相关。结论 Musashi-1、Tcf4、Cdx2可能与贲门腺癌的发生、发展相关,三者在贲门腺癌中表达与组织分化程度有关。远离危险因素可能会减少贲门腺癌的发生。
Objective To study the expression and clinical significance of Musashi-1, Tcf4 and Cdx2 in human gastric cardia adenocarcinoma. Methods The expressions of Musashi-1, Tcf4 and Cdx2 in gastric cardia adenocarcinoma, paracancerous tissues and normal cardia tissues were detected by immunohistochemistry. The expressions of Musashi-1, Tcf4 and Cdx2 in gastric cardia adenocarcinoma and paracancerous tissues were detected by Western blot Expression, and analysis of its expression and clinicopathological features, risk factors. Results The positive expression rates of Musashi-1 in normal cardia epithelium, paracancerous tissue and gastric cardia adenocarcinoma were 20.0%, 45.0% and 79.3% (P <0.05) respectively. The positive rates of Tcf4 were 20.0% and 40.0% The positive expression of Cdx2 was 20.0%, 60.0% and 37.9%, respectively (P <0.05). The results of Western blot showed that the expressions of Musashi-1 and Tcf4 in cancer tissues were higher than those in adjacent tissues and Cdx2 The expression of Musashi-1 and Tcf4 in gastric cardia adenocarcinoma was positively correlated (r = 0.662, P <0.05), while Tcf4 and Cdx2 were weakly correlated in adjacent tissues (r = 0.375 (P <0.05). The expression of Musah-1, Tcf4 and Cdx2 were correlated with the degree of tumor differentiation (P <0.05). Musahi-1, Tcf4, Cdx2 expression and smoking, Helicobacter pylori infection, gastroesophageal reflux disease are closely related. Conclusion Musashi-1, Tcf4, Cdx2 may be related to the occurrence and development of gastric cardia adenocarcinoma. The expression of Musashi-1, Tcf4 and Cdx2 in gastric cardia adenocarcinoma is related to the degree of tissue differentiation. Away from risk factors may reduce the incidence of gastric cardia adenocarcinoma.
