目的观察芹菜素(Apigenin)对小鼠睾丸组织抗氧化酶活性及抗氧化能力的影响,探讨AP对雄性小鼠生殖功能影响机制。方法 200只成年健康SPF级昆明种雄性小鼠按体重随机分为4组:对照组(生理盐水10ml/kg)、低剂量组(AP 252mg/kg)、中剂量组(AP 504mg/kg)和高剂量组(AP 1008mg/kg),每组50只。连续灌胃7、14、21、28、35天,分批处死,取出睾丸,制备匀浆,测定总超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)活力及酰胺腺嘌呤二核苷酸磷酸(NADPH)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、氧化型谷胱甘肽(GSSG)的浓度。结果 7天CAT活力低、中、高剂量组均高于对照组,其中低、中剂量组与对照组差异有统计学意义(P<0.05)。GSH-PX活力,T-SOD活力、MDA浓度及GSH/GSSG在不同时间观察点,各剂量组与同期对照组差异均无统计学意义。T-SOD活力随时间变化总体升高,MDA浓度随时间变化总体降低,GSH/GSSG高剂量组在不同时间观察点均降低。结论本实验条件下,AP可致小鼠睾丸组织呈现氧化应激状态,对小鼠睾丸具有一定的亚急性氧化损伤作用。
Objective To observe the effect of Apigenin on antioxidant enzyme activity and antioxidant capacity in testis of mice and explore the mechanism of AP on reproductive function in male mice. Methods Male adult Kunming SPF Kunming mice were randomly divided into 4 groups according to body weight: control group (10ml / kg normal saline, AP 252mg / kg), middle dose group (504mg / kg) and High dose group (AP 1008mg / kg), each group of 50. The rats were sacrificed at 7, 14, 21, 28 and 35 days. The testes were removed and homogenized. The contents of total superoxide dismutase (SOD), catalase (CAT) The activity of peroxidase (GSH-PX) and the activity of NADPH, MDA, GSH, GSSG concentration. Results The CAT activity in the low, medium and high dose groups was higher than that in the control group on the 7th day. The difference between the low and medium dose groups and the control group was statistically significant (P <0.05). GSH-PX activity, T-SOD activity, MDA concentration and GSH / GSSG were observed at different time points. There was no significant difference between each dose group and the control group at the same period. T-SOD activity increased with time, MDA concentration decreased with time, GSH / GSSG high-dose group at different time points were reduced. Conclusion Under the experimental conditions, AP can cause oxidative stress in the testis of mice, and have some sub-acute oxidative damage on mouse testes.
