目的对具有抗肿瘤活性的茯苓多糖进行提取、纯化以及结构分析。方法采用水提醇沉法提取茯苓多糖,三氯乙酸法除蛋白,凝胶色谱柱纯化得到抗肿瘤活性茯苓多糖ATPCP,高效液相凝胶色谱法测其相对分子质量,样品水解后经过TLC及GC-MS分析确定单糖组成,通过FT-IR、13C-NMR和1H-NMR对其进行结构分析。结果 GPC结果显示抗肿瘤活性茯苓多糖的重均相对分子质量为16 850,多分散性为1.34,TLC及GC-MS结果显示其含有岩藻糖、半乳糖、葡萄糖、甘露糖,各单糖的物质的量比为1∶2.36∶5.49∶2.34。FT-IR和NMR结果显示ATPCP含有α-型和β-型吡喃糖,其主链为1,6-糖苷键,并存在1,4-糖苷键支链。结论茯苓中具有抗肿瘤活性的茯苓多糖是由4种确定的单糖组成并伴有支链的杂多糖。
Objective To extract, purify and structure the tuckahoe polysaccharide with anti-tumor activity. Methods The polysaccharide was extracted by water extraction and alcohol precipitation, the protein was removed by trichloroacetic acid method, and the anti-tumor active polysaccharide ATPCP was purified by gel column chromatography. The relative molecular mass was determined by high performance liquid chromatography. The monosaccharide composition was confirmed by GC-MS analysis, and its structure was analyzed by FT-IR, 13C-NMR and 1H-NMR. Results The GPC results showed that the weight average molecular weight of polysaccharides was 16 850 and the polydispersity was 1.34. The results of TLC and GC-MS showed that the content of fucose, galactose, glucose, mannose, and monosaccharides The ratio of substances is 1: 2.36: 5.49: 2.34. FT-IR and NMR results showed that ATPCP contains α-type and β-type pyranose, the main chain is 1,6-glycosidic bond, and 1,4-glycosidic bond exists. Conclusion Poria polysaccharide with anti-tumor activity is composed of four kinds of defined monosaccharides and branched-chain heteropolysaccharide.
