目的:筛选石斛明目胶囊的水煎醇沉最佳工艺条件。方法:高效液相色谱法测定水煎液和醇沉液中芍药苷的含量;采用正交设计法考察加水量、浸泡时间、煎煮次数、煎煮时间四因素对水煎工艺提取成分提取率的影响;考察醇沉前水煎液相对密度、醇沉浓度、醇沉时间三因素对醇沉工艺有效成分含量的影响。结果:煎煮次数、煎煮时间对水煎工艺有显著影响(P<0.05);醇沉前水煎液相对密度醇沉工艺有显著影响(P<0.05)。结论:结合生产实际,确定石斛明目胶囊水煎最佳工艺是药材不浸泡,第1次煎煮用9倍量水,第2次煎煮用8倍量水,煎煮2次,每次1h;醇沉工艺最佳条件为即煎煮液浓缩密度为1.10~1.15,加入乙醇使含醇量为70%,醇沉时间为12h。
Objective: To screen the optimum conditions for decoction and alcohol precipitation of Shiji Mingmu capsule. Methods: The content of paeoniflorin in water decoction and alcohol decoction was determined by high performance liquid chromatography. The orthogonal design method was used to examine the extraction rate of water extract, extraction time, decocting time and decocting time. The influence of the three factors of the relative density of alcohol decoction, alcohol concentration, and alcohol precipitation time on the content of active ingredients in the alcohol precipitation process was investigated. Results: The number of decocting and decocting time had a significant effect on the decoction process (P<0.05). The relative density and decocting process of decoction before dehydration had a significant effect (P<0.05). Conclusion: Combining with the actual production, it is determined that the best process for decocting the Shiji Mingmu capsules is not soaking the medicinal materials, using 9 times the amount of water for the first boiling, 8 times the amount of water for the second boiling, and boiling twice. 1h; The optimal conditions for the alcohol precipitation process are that the concentration of the decoction liquid is 1.10 to 1.15, the alcohol content is 70%, and the alcohol precipitation time is 12 hours.
