目的:优化人心房组织膜蛋白提取方法。方法:分步离心法提取心肌膜蛋白,SK2膜蛋白用western-blot检测。结果:BCA检测出蛋白浓度,western-blot检测出蛋白条带。结论:此方法能够提取人心房组织的膜蛋白SK2,该方法简单,稳定,可靠,提取的膜蛋白质量好,浓度高,并能进行下一步的western-blot的实验,以便将来检测SK2膜蛋白在窦性和房颤患者心房组织之间表达的差异。
Objective: To optimize the extraction method of human atrial tissue membrane protein. Methods: Myocardial membrane protein was extracted by stepwise centrifugation. The protein of SK2 was detected by western-blot. Results: Protein concentration was detected by BCA and protein band was detected by western-blot. CONCLUSION: This method can extract membrane protein SK2 from human atrial tissue. The method is simple, stable and reliable. The extracted membrane protein has good quality and high concentration, and can carry out the next western-blot experiment to detect the SK2 membrane protein Differences in the expression of atrial tissue between patients with sinus and atrial fibrillation.
