目的了解高血糖对SD大鼠载脂蛋白M(apoM)表达和分泌的影响。方法于大鼠尾静脉置管,采用微量注射泵以2.5 ml/h速度连续6 h泵入25%葡萄糖注射液的方法建立生理性高血糖大鼠模型。对照组采用同样方法泵入生理盐水。Dot-blotting法检测血清apoM浓度的变化,Real-Time PCR法检测肝脏apoM mRNA的表达。采用Mann-Whitney检验进行统计分析。结果高糖组大鼠肝脏apoM mRNA的表达较对照组降低54%(P<0.01),血清apoM的浓度较对照下降32.1%,但差异无统计学意义(P>0.05)。结论高血糖可抑制大鼠载脂蛋白M的表达和分泌。
Objective To investigate the effect of hyperglycemia on the expression and secretion of apolipoprotein M (apoM) in SD rats. Methods The rat model of physiological hyperglycemia was established by injecting the tail vein into the tail vein of rats by means of a micro-injection pump and pumping 25% glucose injection at 2.5 ml / h for 6 h continuously. Control group using the same method of pumping saline. The changes of apoM concentration in serum were detected by Dot-blotting method. The expression of apoM mRNA in liver was detected by Real-Time PCR. Mann-Whitney test for statistical analysis. Results Compared with the control group, the expression of apoM mRNA in the high glucose group was decreased by 54% (P <0.01) and the serum apoM concentration was 32.1% lower than that in the control group, but the difference was not statistically significant (P> 0.05). Conclusion Hyperglycemia can inhibit the expression and secretion of apolipoprotein M in rats.
