目的研究促性腺激素释放激素(GnRH)激动剂(GnRH agonist,GnRHa)对In-R1-G9细胞内胰升血糖素表达的调控作用。方法分别用终浓度为0.1 nmol/L、10 nmol/L、1000 nmol/L的GnRHa处理培养的In-R1-G9细胞2 h(对照组用生理盐水处理),通过Western blot和荧光定量PCR技术观察胰升血糖素在蛋白水平及转录水平的变化。结果与对照组相比,低浓度的GnRHa(0.1nmol/L)使胰升血糖素在蛋白水平和转录水平的表达均有所升高,但差异无统计学意义(P>0.05);而高浓度的GnRHa(10 nmol/L、1000 nmol/L)能明显增强胰升血糖素的表达(P<0.05或P<0.01)。结论一定浓度的GnRHa能增强In-R1-G9细胞内胰升血糖素的表达。
Objective To investigate the regulatory effect of GnRH agonist (GnRH agonist) on glucagon expression in In-R1-G9 cells. Methods The cultured In-R1-G9 cells were treated with GnRHa of 0.1 nmol / L, 10 nmol / L and 1000 nmol / L for 2 h, respectively. The control group was treated with normal saline. Western Blot and quantitative PCR Observe the changes of glucagon at the protein level and transcription level. Results Compared with the control group, low concentration of GnRHa (0.1 nmol / L) increased the expression of glucagon at the protein level and transcription level, but the difference was not statistically significant (P> 0.05) Concentration of GnRHa (10 nmol / L, 1000 nmol / L) significantly increased the expression of glucagon (P <0.05 or P <0.01). Conclusion A certain concentration of GnRHa can enhance the expression of glucagon in In-R1-G9 cells.
