目的了解基质金属蛋白酶抑制剂对肝癌生长和侵袭转移的影响及其机理。方法通过腹腔注射BB-94对原位移植的转移性人肝癌棵鼠模型LCI-D20进行治疗,观察BB-94对肝癌生长和侵袭转移等影响,并通过明胶酶谱法、反转录聚合酶链反应(RT-PCR)、体外侵袭试验、MTT试验等方法对BB-94对肝癌生长和侵袭转移等的影响的机理进行了研究。结果BB-94对LCI-D20肿瘤生长、侵袭转移均有明显的抑制作用,并能延长其生存期;LCI-D20组织中可见基质金属蛋白酶1、2、9的表达;BB94可抑制肝癌细胞林SMMC-7721的体外侵袭,但不能抑制SMMC-7721的增殖。结论BB-94可抑制肝细胞癌的生长和侵袭转移。
Objective To investigate the effect and mechanism of matrix metalloproteinase inhibitors on the growth, invasion and metastasis of hepatocellular carcinoma. METHODS: LCI-D20, a metastatic human hepatoma model transplanted orthotopically was treated by intraperitoneal injection of BB-94. The effects of BB-94 on hepatocellular carcinoma growth, invasion and metastasis were observed. Gelatin zymography and reverse transcriptase polymerase were used. The mechanism of the effect of BB-94 on the growth, invasion and metastasis of hepatocellular carcinoma was studied by means of chain reaction (RT-PCR), in vitro invasion assay, and MTT assay. Results BB-94 significantly inhibited the growth, invasion and metastasis of LCI-D20 tumors, and prolonged its survival. The expression of matrix metalloproteinases 1,2,9 was observed in LCI-D20 tissues. BB94 inhibited the growth of hepatocellular carcinoma cells. In vitro invasion of SMMC-7721 did not inhibit the proliferation of SMMC-7721. Conclusion BB-94 can inhibit the growth, invasion and metastasis of hepatocellular carcinoma.
