目的检测趋化因子CXCL10及其受体CXCR3在幼年特发性关节炎(JIA)患儿关节滑膜组织中的表达,探讨其在JIA发病机制中的作用。方法通过免疫组织化学方法,检测12例JIA患儿和4例非JIA患儿关节滑膜组织中CXCL10和CXCR3的表达;通过半定量RT-PCR方法,检测CXCR3在JIA患儿和对照幼儿关节滑膜组织中mRNA水平的表达。结果免疫组织化学结果表明,CXCL10/CXCR3在JIA患儿及对照组阳性表达差异有显著性(P<0.05);CXCR3在JIA患儿关节滑膜组织中mRNA表达水平(CXCR3∶GAPDH为2.26±1.55)显著高于对照组(0.66±0.44),两者差异有显著性(P<0.05)。结论趋化因子CXCL10及其受体CXCR3在幼年特发性关节炎(JIA)的发病中可能起到了重要的作用。
Objective To investigate the expression of chemokine CXCL10 and its receptor CXCR3 in synovial tissue of juvenile patients with idiopathic arthritis (JIA), and to explore its role in the pathogenesis of JIA. Methods The expression of CXCL10 and CXCR3 in synovial tissues from 12 JIA children and 4 non-JIA children was detected by immunohistochemistry. The CXCR3 expression was detected by semi-quantitative RT-PCR in children with JIA and controls Membrane tissue expression of mRNA levels. Results The results of immunohistochemistry showed that the expression of CXCL10 / CXCR3 in JIA children and controls was significantly different (P <0.05). The expression of CXCR3 in synovial tissues of JIA children (CXCR3:GAPDH was 2.26 ± 1.55 ) Was significantly higher than the control group (0.66 ± 0.44), the difference was significant (P <0.05).
