从细胞形态学和细胞酶组织化学的角度出发,用伪足法和NBT 法检测体外中性粒细胞自发性激活,研究了抗凝剂、孵育温度和性别对检测结果的影响。发现:(1)EDTA 可显著抑制中性粒细胞自发性激活,血标本以小剂量肝素抗凝为宜;(2)孵育温度对中性粒细胞自发性激活有影响,37 ℃比25 ℃的激活百分率高,最高激活百分率出现较25 ℃早1 h;(3)女性中性粒细胞自发性激活百分率较男性高,最高激活百分率出现较男性早1 h。研究结果为以后深入研究中性粒细胞激活与多种疾病的关系提供了参照系统。
The effects of anticoagulants, incubation temperature and sex on the test results were studied by using pseudopod method and NBT method to detect the spontaneous neutrophil activation in vitro from the aspects of cell morphology and enzymatic histochemistry. It is found that: (1) EDTA can significantly inhibit the spontaneous activation of neutrophils, blood samples with low-dose heparin anticoagulation is appropriate; (2) incubation temperature on the spontaneous activation of neutrophils, 37 ℃ than 25 ℃ The percentage of activation was higher than that of 25 ℃. (3) The percentage of spontaneous activation of female neutrophils was higher than that of men, the highest percentage of activation was 1 h earlier than that of male. The results provide a reference system for further study on the relationship between neutrophil activation and various diseases.
