目的:观察脂肪间充质干细胞(ADMSCs)心肌内移植对兔心肌梗死(MI)后心力衰竭近期心功能的影响,并初步探讨其机制。方法:将30只兔随机分为假手术组(只穿线不接扎)、对照组(心肌内注射IMDM培养基)及干细胞移植组(心肌内注射ADMSCs),每组10只(n=10);结扎兔冠状动脉左前降支(LAD)制作急性MI模型,同时将6-二氨基-乙-苯茚二酮(DAPI)标记的ADMSCs注射到梗死区数个不同部位。造模前及移植后4周、8周,用超声心动图检测心功能,术后8周处死动物测量左室体质量指数及组织学观察,采用免疫荧光方法鉴定移植的细胞。结果:移植后4周起,心功能指标明显改善(P<0.05),术后第8周心功能指标改善更明显(P<0.01),左室重构程度也得到明显改善(P<0.01)。结论:移植的ADMSCs可在宿主体内成活,并显著改善兔MI后的心功能。
OBJECTIVE: To observe the effect of intramyocardial transplantation of adipose-derived mesenchymal stem cells (ADMSCs) on the recent cardiac function in heart failure after myocardial infarction (MI) in rabbits and to explore its mechanism. Methods: Thirty rabbits were randomly divided into sham-operated group (IMGCs only), control group (intramyocardial injection of IMDM medium) and stem cell transplantation group (intramyocardial injection of ADMSCs), 10 rats in each group (n = 10) The acute MI model was made by ligating the left anterior descending artery (LAD) of rabbits and the ADMSCs labeled with DAPI were injected into several different parts of infarction area. Heart function was measured by echocardiography before and 4 and 8 weeks after transplantation. Left ventricular mass index and histological observation were performed at 8 weeks after transplantation. The transplanted cells were identified by immunofluorescence staining. Results: At 4 weeks after transplantation, cardiac function improved significantly (P <0.05), and cardiac function improved more significantly after 8 weeks (P <0.01) and left ventricular remodeling improved significantly (P <0.01) . CONCLUSION: Transplanted ADMSCs survive in the host and significantly improve cardiac function after rabbit MI.
