目的:建立脂肪乳注射液及卵磷脂中溶血磷脂酰胆碱(LPC)的薄层色谱检查方法。方法:样品用正庚烷-异丙醇(1∶2)溶解后直接点于硅胶G薄层板上;展开剂为氯仿-无水乙醇-三乙胺-水(10∶13∶5∶3),显色剂为Dittmer-Lester试剂;薄层扫描条件:双波长锯齿扫描,测定波长680nm,参比波长400nm,线性参数SX=3,狭缝宽度0.4mm×0.4mm,灵敏度中等。结果:在上述条件下,样品中LPC斑点与其他磷脂斑点分离良好,LPC在0.4~10μg范围内线性良好,r=0.9997,平均加样回收率为99.4%,RSD=2.1%。结论:本法简便、快速、成本低,可用于脂肪乳注射液及卵磷脂中LPC的常规检查。
Objective: To establish a method for the determination of lysophosphatidylcholine (LPC) in fat emulsion injection and lecithin. Methods: The sample was dissolved in n-heptane-isopropanol (1: 2) and directly on silica gel G plate. The developing solvent was chloroform-absolute ethanol-triethylamine- ), The color reagent Dittmer-Lester reagent; TLC scanning conditions: dual wavelength sawtooth scan, the determination wavelength of 680nm, the reference wavelength of 400nm, the linear parameter SX = 3, the slit width of 0.4mm × 0.4mm, moderate sensitivity. Results: Under the above conditions, the LPC spots were well separated from other phospholipid spots. The linear range of LPC was 0.4 ~ 10μg, r = 0.9997, the average recovery was 99.4% and the RSD was 2.1%. Conclusion: This method is simple, rapid and low cost and can be used for the routine examination of LPC in fat emulsion injection and lecithin.
