植物体内生长素输出载体PIN蛋白基因的表达变化间接反映了生长素的运输与积累状态。为分析生长素在狗蔷薇(Rosa canina)类原球茎发生初期的作用,以狗蔷薇叶片愈伤形成的不定根为材料,分离了生长素输出载体蛋白基因PIN1和PIN2的cDNA,分别命名为RcPIN1(GenBank登录号KF543362)和RcPIN2(GenBank登录号KF543363)。RcPIN1全长2 266 bp,编码621个氨基酸;RcPIN2全长2 248 bp,编码643个氨基酸,两者推导蛋白结构差异微小,在N端和C端均有5个跨膜区域,中间1个亲水区。Blast比对发现两个基因与多种植物PIN基因具有高度同源性(>70%)。半定量RT-PCR分析表明,RcPIN1在根、茎中表达量高于叶和花,RcPIN2在根中表达水平高于茎、叶和花;在狗蔷薇类原球茎发生初期,TDZ诱导培养下愈伤—不定根RcPIN1和RcPIN2表达上调,而TDZ+TIBA抑制培养下两基因表达不活跃,且类原球茎形成率降低。试验结果表明生长素的极性运输与积累参与了调控狗蔷薇类原球茎的初期形态建成。
Plant auxin export vector PIN protein gene expression changes indirectly reflect the transport and accumulation of auxin status. In order to analyze the role of auxin in the early development of protocorm-like bodies of Rosa canina, the cDNAs encoding auxin export vector protein PIN1 and PIN2 were isolated from the adventitious roots formed by callus induction of R. roseus and named as RcPIN1 ( GenBank Accession No. KF543362) and RcPIN2 (GenBank Accession No. KF543363). RcPIN1 is 2 266 bp in length and encodes 621 amino acids. RcPIN2 is 2 248 bp in length and encodes 643 amino acids. The deduced amino acid sequence of RcPIN1 has 5 small transmembrane domains and 5 transmembrane domains in the N and C terminus. Water area. The Blast alignment showed that the two genes are highly homologous (> 70%) to multiple plant PIN genes. Semi-quantitative RT-PCR analysis showed that the expression level of RcPIN1 in roots and stems was higher than that in leaves and flowers, and the expression level of RcPIN2 in roots was higher than that in stems, leaves and flowers. Injury - adventitious roots RcPIN1 and RcPIN2 expression was up-regulated, while TDZ + TIBA inhibited the expression of the two genes are not active, and the formation of protocorm-like bodies decreased. The experimental results show that the polar transport and accumulation of auxin are involved in the regulation of the initial morphology of protocorm-like bodies of dog rose.
