在肾脏超微结构研究中采用锇酸加亚铁氰化钾后固定技术,在增强肾组织膜性结构的反差,保存糖元方面均优于锇酸。现报道如下。材料和方法本文采用人肾活检组织作为研究材料。方法:人肾活检组织分切成1 mm~3,3.75%戊二醛固定,磷酸缓冲液漂洗,分三种方法后固定:(1)1%锇酸后固定2小时;(2)2%锇酸与2%亚铁氰化钾等量混合(二者最终浓度均为1%)后固定2小时;(3)1%亚铁氰化钾后固定2小时。后固定完毕均用0.1MPH7.4磷酸缓冲液冲洗,逐级丙酮脱水,
The use of osmium tetroxide plus potassium ferrocyanide posterior fixation in renal ultrastructure studies is superior to osmium tetroxide in enhancing the contrast of membranous structure in renal tissues and preserving glycogen. Report as follows now. Materials and Methods Human renal biopsy tissue was used as the research material. Methods: Human renal biopsy was cut into 1 mm ~ 3. 75% glutaraldehyde and rinsed with phosphate buffer.
