目的探讨重组人粒细胞/巨噬细胞集落刺激因子(rhGM-CSF)对放疗化疗后动物血象等的影响。方法利用家犬和小白鼠分别服用环磷酰胺和60Co照射制造的动物模型,测定注射rhGM-CSF后动物白细胞、血小板、红细胞、血红蛋白、网织细胞、脾集落数、骨髓有核细胞、巨噬细胞及病理组织变化等。结果注射12和24μg/kg剂量的rhGM-CSF显著抑制家犬由环磷酰胺所致白细胞和血小板的减少;注射75、150、300μg/kg剂量rhGM-CSF显著增加60Co照射后小鼠脾细胞集落、外周白细胞、骨髓有核细胞和巨噬细胞。结论rhGM-CSF有明显抑制环磷酰胺及60Co照射所致白细胞减少的作用,且与进口升白能药效相似。
Objective To investigate the effects of recombinant human granulocyte / macrophage colony-stimulating factor (rhGM-CSF) on hemogram of animals after radiotherapy and chemotherapy. Methods Animals and mice were treated with cyclophosphamide and 60Co irradiation respectively. The levels of white blood cells, platelets, erythrocytes, hemoglobin, reticulocytes, spleen colonies, bone marrow nucleated cells, macrophages Cell and pathological changes. Results At doses of 12 and 24 μg / kg, rhGM-CSF significantly inhibited the decrease of white blood cells and platelets induced by cyclophosphamide in dogs. Administration of 75, 150 and 300 μg / kg doses of rhGM-CSF significantly increased splenocyte colonies , Peripheral leukocytes, bone marrow nucleated cells and macrophages. Conclusion rhGM-CSF can significantly inhibit the leukopenia induced by cyclophosphamide and 60Co irradiation, and has similar efficacy to imported lightening.
