将顺铂和磁性氧化铁共包于白蛋白中而制成了磁性靶向顺铂白蛋白微球。按正交设计筛选最佳合成工艺为:顺铂用量为40mg,白蛋白用量为250mg,FeCl2用量为282.8mg,pH=10。并以微球进行了质量考察:微球平均粒径为2.89±0.51μm,磁性靶向顺铂微球的载药量为13.64%(mg/mg),包裹率为85.24%;含药微球在体外12h释放完全,具一定的缓释作用;稳定性考察表明所制微球稳定性良好。
Cisplatin and magnetic iron oxide were co-encapsulated in albumin to make magnetically targeted cisplatin albumin microspheres. The optimal synthesis procedure for screening by orthogonal design was as follows: 40 mg of cisplatin, 250 mg of albumin, 282.8 mg of FeCl2, and pH=10. The microspheres were used to investigate the quality: the average diameter of the microspheres was 2.89±0.51μm, the loading capacity of the magnetic targeting cisplatin microspheres was 13.64% (mg/mg), and the encapsulation rate was 85. 24%; drug-loaded microspheres released completely in vitro at 12h, with a certain degree of sustained release; Stability study showed that the microspheres made of good stability.
