目的 :通过动物实验观察氯沙坦的降压效果及对心肌纤维化的作用。方法 :自发性高血压大鼠 16只随机分为治疗组 (SHR L)和对照组 (SHR C) ,另设同源的WKY大鼠 10只为正常对照组。治疗组口服氯沙坦 2 0mg/kg,qd ,服药 6周后处死动物 ,取左心室心肌称重 ,计算左室 /体重比 (LVW/BW) ,Van Gieson染色观察左心室心肌胶原变化 ,全自动图像分析系统测量心肌间质胶原容积百分比 (CVF)和血管周围胶原面积 /周长比(PVCA/PCL)。结果 :治疗组服药后血压明显下降 ,服药前后心率无明显变化 ,左室心肌LVW /BW ,CVF及PV CA/PCL服药后明显下降。结论 :氯沙坦对自发性高血压大鼠治疗 6周可以有效降低血压 ,部分逆转心肌纤维化和左室肥厚 ,对心率无明显影响
OBJECTIVE: To observe the antihypertensive efficacy of losartan and its effect on myocardial fibrosis through animal experiments. Methods: Twenty-six spontaneously hypertensive rats were randomly divided into treatment group (SHR L) and control group (SHR C), and 10 WKY rats were used as control group. The rats in the treatment group were given Losartan 20 mg / kg, qd for 6 weeks, and the animals were sacrificed. The left ventricular myocardium was weighed and the left ventricular mass ratio (LVW / BW) was calculated. The changes of collagen in left ventricular myocardium were observed by Van Gieson staining. Automated image analysis system measures myocardial interstitial collagen volume fraction (CVF) and perivascular collagen area / perimeter ratio (PVCA / PCL). Results: After treatment, the blood pressure of the treatment group decreased significantly. The heart rate did not change before and after taking the medicine. The LVW / BW, CVF and PV CA / PCL of the left ventricle decreased obviously after taking the medicine. Conclusion: Losartan on spontaneous hypertensive rats for 6 weeks can effectively reduce blood pressure, partial reversal of myocardial fibrosis and left ventricular hypertrophy, no significant effect on heart rate
