目的:探讨激素替代治疗(HRT)对宫颈腺癌预后的影响。方法:①体外培养人宫颈癌Hela细胞,采用四甲基偶氮唑蓝(MTT)比色法检测两种激素对Hela细胞增殖的影响;流式细胞仪(FCM)检测细胞生长周期和凋亡率的影响;免疫组化方法测定其雌激素受体(ER)和孕激素受体(PR)的表达。②对24例宫颈腺癌治疗后的妇女给予HRT,通过患者主观症状的变化了解替代效果;通过阴道宫颈涂片、B超、胸片、ECT等检查了解病情变化,最终总结其疗效及对预后的影响,判断其可行性。结果:①雌二醇(E2)+孕酮(P)对Hela细胞生长有抑制作用(P<0.05);E2+P组Hela细胞S期细胞比例下降,凋亡率上升;E2和P对人宫颈腺癌细胞ER和PR的表达无显著影响。②复发情况:研究组中复发转移占29.17%(7/24),对照组中复发转移占40.00%(8/20),两组差异无统计学意义(P>0.05)。结论:E2对宫颈癌Hela细胞具有促进生长,抗凋亡作用,而P抑制其生长,诱导凋亡,P能有效抵消E2对Hela细胞生长的刺激作用。宫颈腺癌患者的复发与病理类型、临床分期有密切关系,与是否采用HRT关系不大。
Objective: To investigate the effect of hormone replacement therapy (HRT) on the prognosis of cervical adenocarcinoma. Methods: ① Hela cells were cultured in vitro. The effects of two hormones on the proliferation of Hela cells were detected by MTT assay. The growth and apoptosis of Hela cells were detected by flow cytometry (FCM) The expression of estrogen receptor (ER) and progesterone receptor (PR) was determined by immunohistochemistry. ②Twenty-four patients with cervical adenocarcinoma were treated with HRT, and the subjective symptoms of patients were analyzed to understand the effect of replacement. The vaginal smear, B-ultrasound, chest radiograph, ECT and other tests were used to understand the changes of the disease. There was no significant difference between the two groups (P> 0.05). Conclusion: E2 can promote the growth and anti-apoptotic effect of cervical cancer Hela cells, while P inhibits its growth and induces apoptosis. P can effectively counteract the stimulatory effect of E2 on the growth of Hela cells.
