本文报道了利用PCR技术检测了34例丙型肝炎病毒E2/NS1基因的cDNA并与5’非编码区基因的cDNA检测技术进行了比较,应用pUC18质粒对此扩增片段进行了克隆与序列分析,结果表明克隆片段HCV-S1为我国主要流行的HCV基围亚型-II型,其与HCV-II型的核苷酸与氨基酸的同源性均在90%以上,序列分析表明该片段富含脯氨酸和胱氨酸,可能具有复杂的空间构型,且与4个糖基化位点一样保持稳定,另外,在HCV-S1片段的3’端有一36个核苷酸的区域,其变化可能具有型特异性
In this paper, we report the detection of cDNA of 34 cases of hepatitis C virus E2 / NS1 gene by PCR and compare with the cDNA of 5 ’non-coding region. The pUC18 plasmid was cloned and sequenced The results showed that the cloned fragment of HCV-S1 is the most popular HCV subtype-II in our country, and its nucleotide and amino acid homology with HCV-II is over 90%.
