目的探讨藏药翼首草总苷(TGP)对佐剂性关节炎(AA)大鼠的治疗作用及其作用机制。方法取健康SD大鼠48只,随机选取8只作为正常组,其余40只大鼠右足趾皮内注射0.1 m L完全弗氏佐剂(CFA),致炎17 d后,随机分为5组,每组8只,即模型组,醋酸地塞米松组(0.001 g·kg-)1,翼首草总苷低、中、高剂量组(0.058,0.116,0.232 g·kg-)1,连续灌胃给药30 d。观测致炎后17,27,37,47 d大鼠足趾肿胀率、苏木素-伊红(HE)染色法观察致炎侧踝关节滑膜病理改变。致炎47 d,ELISA法测定血清中白介素-1β(IL-1β)、白介素-17(IL-17)、肿瘤坏死因子-α(TNF-α)以及膝关节滑膜核因子-κB(NF-κB)的水平。结果与正常组比较,模型组足趾肿胀率明显升高(P<0.01),滑膜细胞、巨噬细胞、纤维细胞大量增生,炎症细胞大量浸润,而且IL-1β、TNF-α、IL-17、NF-κB水平明显升高(P<0.01)。与模型组比较,各给药组足趾肿胀率明显降低(P<0.05,P<0.01),并能改善滑膜细胞、巨噬细胞、纤维细胞增生与炎症细胞浸润,IL-1β、TNF-α、IL-17、NF-κB水平也明显降低(P<0.05,P<0.01)。结论翼首草总苷能抑制AA大鼠关节肿胀,改善AA大鼠踝关节滑膜病变,对佐剂性关节炎大鼠具有治疗作用,其作用机制可能与降低体内IL-1β、TNF-α、IL-17、NF-κB的水平有关。
Objective To investigate the therapeutic effect and its mechanism of Tibetan medicinal herb TGP on adjuvant arthritis (AA) rats. Methods Totally 48 healthy SD rats were randomly divided into normal group (n = 8) and 0.1 m L complete Freund’s adjuvant (CFA) injected into the right foot of the right foot. After 17 days of inflammation, they were randomly divided into 5 groups , Each group of 8, namely, model group, dexamethasone acetate group (0.001 g · kg-) 1, Pterocaryosides low, middle and high dose group (0.058,0.116,0.232 g · kg-) 1, continuous Gavage for 30 days. The swelling rate of toes was observed on 17, 27, 37, and 47 days after inflammation in rats. The pathological changes of the ankle joint synovium were observed by hematoxylin-eosin staining. The serum levels of IL-1β, IL-17, TNF-α and NF- κB) levels. Results Compared with the normal group, the swelling rate of the toe in the model group was significantly increased (P <0.01), the hyperplasia of synovial cells, macrophages and fibrocytes, and the infiltration of inflammatory cells were significant. The levels of IL-1β, TNF- 17, NF-κB levels were significantly increased (P <0.01). Compared with the model group, the swelling rate of toes in each group was significantly decreased (P <0.05, P <0.01), and could improve the proliferation of synovial cells, macrophages, fibroblasts and inflammatory cells, IL- α, IL-17, NF-κB levels were significantly lower (P <0.05, P <0.01). Conclusion The total glycosides of Pterygota can inhibit the swelling of joints in AA rats and improve the ankle synovial lesions of AA rats, which may have the therapeutic effect on adjuvant arthritis rats. The mechanism may be related to the decrease of IL-1β, TNF-α , IL-17, NF-κB levels.
