目的:探讨2种肝癌细胞BEL-7402细胞及BEL-7402/FU细胞中差异表达的miRNA并对其基因表达谱进行生物学信息分析。方法:采用TRIzol一步法提取BEL-7402细胞及BEL-7402/FU细胞的总RNA,并纯化mRNA,反转录合成荧光分子Hy3标记的cDNA探针,与基因芯片杂交;采用Genepix Pro 6.0图像分析软件对芯片图像进行分析,把图像信号转化为数字信号,然后以差异为2倍的标准来确定差异表达基因。结果:在333个基因表达谱的筛选中,发现有2个基因表达水平显著上调,331个基因表达水平显著下调。结论:miR-122,miR-195等基因组对表达肝癌细胞耐药基因表达谱有显著的影响,可能参与肝癌耐药的发生、发展。
OBJECTIVE: To explore differentially expressed miRNAs in two hepatocellular carcinoma cell lines, BEL-7402 and BEL-7402 / FU cells, and to analyze their gene expression profiles for biological information. Methods: The total RNA of BEL-7402 cells and BEL-7402 / FU cells was extracted by one-step TRIzol method. The mRNA of purified Bcl-7402 / FU cells was reverse transcribed and hybridized with Hy3 labeled cDNA probe. Genepix Pro 6.0 image analysis The software analyzes the image of the chip, converts the image signal into a digital signal, and then determines differentially expressed genes by a factor of two. Results: In the 333 gene expression profiling, two gene expression levels were found to be significantly up-regulated and 331 genes were significantly down-regulated. Conclusion: The miR-122, miR-195 and other genomes have significant effects on the expression of drug resistance gene expression in hepatocellular carcinoma cells, which may be involved in the occurrence and development of drug resistance in hepatocellular carcinoma.
