目的调查柳州地区献血人群中HBV的基因型分布和不同基因型HBV的群体动力学。方法采集来自广西血液中心2009年HBV确证阳性的献血者血浆标本,用半巢式PCR的方法扩增HBV基因组S区的序列,测序后通过贝叶斯推断探讨柳州地区分布的HBV基因型;应用系统发育学和群体遗传学方法,对不同的HBV基因型群体进行群体遗传学分析,探讨该地区不同基因型HBV的扩张情况。结果柳州地区献血者中HBV有3种基因型,分别是B型(71.4%)、C型(26.5%)和D型(2%)。B型和C型HBV群体的Tajima’s D检验与Fu’s Fs检验结果都为负值,同时这2个群体的错配分布也呈现单一峰倒钟形分布,说明柳州地区的HBV群体都在扩张。贝叶斯轮廓图推断B型和C型HBV目前有相似的有效种群大小,但起源时间B型晚于C型。结论柳州地区献血者中HBV基因型多样性较丰富,B型和C型的HBV都正在经历1个缓慢扩张的过程,需持续研究和监控HBV在献血者中的感染情况和遗传分化情况,做好相应的血液预警工作。
Objective To investigate the distribution of HBV genotypes and the population dynamics of different genotypes of HBV in blood donors in Liuzhou area. Methods Plasma samples from blood donors with HBV positive confirmation from Guangxi Hematology Center in 2009 were collected. The sequence of HBV S region was amplified by semi-nested PCR and sequenced. The genotypes of HBV in Liuzhou were determined by Bayesian inference. Phylogenetics and population genetics methods, population genetic analysis of different HBV genotypes to explore the expansion of different genotypes of HBV in the region. Results There were three genotypes of HBV in blood donors in Liuzhou area, which were type B (71.4%), type C (26.5%) and type D (2%) respectively. The Tajima’s D test and Fu’s Fs test results of HBV genotypes B and C were both negative, and the distribution of mismatch also showed a single peak and bell-shaped distribution, indicating that HBV population in Liuzhou area was expanding. Bayesian profiles infer that HBV types B and C presently have similar effective population sizes, but the origin of time type B is later than that of type C. Conclusion There are abundant HBV genotypes in blood donors in Liuzhou area. Both HBV genotypes B and C are experiencing a slowly expanding process.
