目的观察MMP-2和MMP-9在肺组织损伤修复过程中的表达和变化规律,并探讨它们在肺组织损伤修复中作用的分子机制和意义。方法20只Wister大白鼠,随机分为一个正常组和臭氧攻击损伤后不同时间的实验组,制作大鼠肺损伤模型。应用免疫组化法和图像分析技术观察各组MMP-2及MMP-9蛋白表达的强度。结果成功复制大鼠肺组织损伤动物模型;实验观察表明,MMP-2、MMP-9蛋白表达在正常肺组织为弱阳性,在肺组织损伤时,表达上调,且随着肺组织损伤修复行进而先逐渐增强(<0.05),继而逐渐减弱(<0.05),当肺组织损伤修复达正常时,MMP-2、MMP-9表达恢复到正常水平(>0.05)。结论MMP-2、MMP-9的高表达与肺组织损伤密切相关。
Objective To observe the expression of MMP-2 and MMP-9 in the process of lung injury and repair, and to explore the molecular mechanism and significance of their roles in the repair of lung injury. Methods Twenty Wister rats were randomly divided into a normal group and experimental groups at different time points after ozone challenge. Rat models of lung injury were made. The intensity of MMP-2 and MMP-9 protein expression in each group was observed by immunohistochemistry and image analysis. Results The rat model of lung injury was successfully established. The experimental results showed that the expressions of MMP-2 and MMP-9 were weakly positive in normal lung tissue and upregulated in lung tissue, and with the progression of lung injury (P <0.05), and then gradually weakened (P <0.05). The expression of MMP-2 and MMP-9 returned to normal level (> 0.05) when the lung tissue repaired to normal. Conclusion The high expressions of MMP-2 and MMP-9 are closely related to lung injury.
