奎尼丁因减低地高辛的肾清除和非肾清除以及降低地高辛分布容积而升高地高辛血清浓度,曾有导致地高辛中毒的报道。虽然安体舒通也有报告升高地高辛血清水平,但其临床意义尚未充分阐明。本文研究奎尼丁和安体舒通联用对地高辛药物动力学的影响。受试者为男女各三名,身体健康。血清肌酐,尿素氮,胆红素,谷-草转氨酶(GOT),碱性磷酸酶均正常;并未用其它药物。实验共分四期,至少隔3周。各为(1)地高辛1mg10分钟内静注。(2)地高辛静注1mg,加注射前三天至注后五天持续口服给予硫酸奎尼丁每6小时200mg。(3)地高辛1mg静注,注射前三天至注后五天持续每6小时口服安体舒通50mg。(4)三种药物联合使
Quinidine has been reported to cause digoxin poisoning because digoxin reduces renal clearance and non-renal clearance of digoxin, and decreases digoxin levels. Although spironolactone has also been reported to increase digoxin serum levels, its clinical significance has not yet been fully elucidated. This article studies the effect of quinidine and spironolactone on digoxin pharmacokinetics. Subjects for men and women of three, in good health. Serum creatinine, urea nitrogen, bilirubin, Valley - grass aminotransferase (GOT), alkaline phosphatase were normal; no other drugs. The experiment is divided into four phases, at least every 3 weeks. Each (1) digoxin 1mg10 minutes intravenous injection. (2) intravenous injection of digoxin 1mg, plus three days before injection to five days after the injection of oral administration of quinidine sulphate 200mg every 6 hours. (3) digoxin 1mg intravenous injection of three days before injection to five days after the injection of spironolactone every 6 hours 50mg. (4) three kinds of drugs combined to make
