目的:探究益气温阳补肾法对甲减模型大鼠血清TNF-α、IFN-γ的调节作用。方法:60只Wistar大鼠,随机抽取10只为空白组,雌雄各半,其余50只予0.1%PTU生理盐水溶液灌胃建造大鼠甲减模型。15天后检测甲功,FT3、FT4水平低于空白组,TSH>10u IU/ml为造模成功。将50只甲减模型大鼠随机分为模型组、西药组、中药低剂量组、中药中剂量组、中药高剂量组,每组10只,雌雄各半,予相应药物治疗12周后,腹主动脉取血,检测血清FT3、FT4、TSH、TNF-α、IFN-γ水平。结果:治疗后,与模型组相比,各治疗组FT3、FT4水平上升,TSH水平下降(p<0.05),仅中药中、高剂量组TNF-α水平低于模型组(p<0.05),西药及中药各剂量组IFN-γ水平低于模型组(p<0.05),但中药中、高剂量组IFN-γ水平较西药组更低(p<0.05)。结论:益气温阳补肾法可以改善甲减模型大鼠的甲状腺功能,调节TNF-α、IFN-γ水平,但需要较高剂量。
Objective: To investigate the regulation effect of Yiqi Wenyang Bushen Method on serum TNF-α and IFN-γ in rats with hypothyroidism. Methods: Totally 60 Wistar rats were randomly divided into blank control group (10 rats) and male and female mice (half male and half female). The remaining 50 rabbits were given 0.1% PTU saline solution to establish model of hypothyroidism. 15 days after the detection of thyroid function, FT3, FT4 levels lower than the blank group, TSH> 10u IU / ml for modeling success. Fifty hypothyroid model rats were randomly divided into model group, western medicine group, low dose Chinese medicine group, middle dose Chinese medicine group and traditional Chinese medicine high dose group, with 10 rats in each group. Aorta blood, serum FT3, FT4, TSH, TNF-α, IFN-γ levels. Results: Compared with the model group, the levels of FT3 and FT4 and the levels of TSH in each treatment group decreased (p <0.05), and only the levels of TNF-α in the medium and high dose groups were lower than those in the untreated group (p <0.05) The levels of IFN-γ in western medicine and traditional Chinese medicine groups were lower than those in model group (p <0.05), but the levels of IFN-γ in middle and high dose groups were lower than those in western medicine group (p <0.05). Conclusion: Yiqi Wenyang Bushen Method can improve thyroid function, regulate the levels of TNF-α and IFN-γ in hypothyroid model rats, but need higher dose.
