目的:探讨巢式RT-PCR在急性髓系白血病(AML)融合基因检测中的应用价值。方法:收集2008年1月-2010年1月在我院初治的227例AML患者的骨髓细胞,提取RNA转录cDNA后,以多重巢式RT-PCR进行融合基因检测。结果:227例患者中共检出融合基因95例(41.9%),包括AML1/ETO,CBFβ/MYH11,MLL基因重排,PML/RARα,CBFβ/MYH11,PLZF/RARα,MLL/AF6,BCR/ABL。结论:巢式RT-PCR可以较为快速准确的对融合基因进行检测,为AML的诊断和治疗提供依据。
Objective: To explore the value of nested RT-PCR in the detection of acute myeloid leukemia (AML) fusion gene. Methods: Bone marrow cells from 227 patients with AML treated in our hospital from January 2008 to January 2010 were collected. RNA was extracted and transcribed into cDNA. Multiplex nested RT-PCR was used to detect the fusion gene. RESULTS: A total of 95 (41.9%) fusion genes were detected in 227 patients, including AML1 / ETO, CBFβ / MYH11, MLL gene rearrangement, PML / RARα, CBFβ / MYH11, PLZF / RARα, MLL / AF6, BCR / ABL . Conclusion: The nested RT-PCR can detect the fusion gene more quickly and accurately, and provide the basis for the diagnosis and treatment of AML.
