作者应用纸上色层法和末端基团分析法,分析分枝杆菌素水解液中短肽链的氨基酸以及N-末端氨基酸时,阐明了氨基酸排列顺序。方法:取本品盐酸水解液,用双向纸上层析法进行肽的分离,测得水解液中短肽,然后进行洗脱,把每个短肽各别再次盐酸水解,水解液经纸上层析,即能得出氨基酸在短肽中排列顺序。同时进行亚硝酰氯法的脱氨基作用和二硝基苯氟(DNP)法测定N-末端氨基在肽链中位置。
The authors applied paper chromatography and end-group analysis to elucidate the amino acid sequence of the short chain amino acids and the N-terminal amino acids in mycobactin hydrolyzate. Methods: Hydrochloric acid hydrolyzate of this product was taken and the peptide was separated by two-way paper chromatography. The short peptide in the hydrolyzate was measured and then eluted. Each of the short peptides was hydrolyzed again by hydrochloric acid. The hydrolyzate was passed through the paper Chromatography, which can be drawn amino acids in the short peptide in the order. At the same time, the deamination of nitrosyl chloride method and dinitrofluorobenzene (DNP) method were used to determine the position of N-terminal amino group in the peptide chain.
