目的研究六价铬暴露对职业人群细胞周期相关基因表达的影响。方法选取某电镀厂155名六价铬职业暴露工人为研究对象,调查工人基本情况并采集肝素抗凝外周血,采用电感耦合等离子体质谱仪检测血铬水平(ppb),根据血铬水平将研究对象分为4组(组1≤0.04 ppb、0.04 ppb<组2≤1 ppb、1 ppb<组3≤5 ppb、组4>5 ppb),比较各组p16和CDK6基因表达水平,并分析基因表达的影响因素。结果共纳入155名工人,其中男性89人,女性66人,平均(39.65±8.86)岁,工龄中位数为36个月,血铬中位数为0.95 ppb。4组p16基因表达水平中位数(四分位间距)分别为4.22(19.91)、7.19(30.71)、7.47(21.47)和14.60(27.28),组间差异无统计学意义(P=0.19)。4组CDK6基因表达水平中位数(四分位间距)分别为4.00(8.13)、15.05(32.72)、8.03(17.32)和24.81(35.09),其中组1的CDK6基因表达水平低于其他各组(P<0.01)。Spearman相关分析结果显示,CDK6基因表达水平与血铬水平呈正相关(rs=0.28,P<0.01);Logistic回归分析结果显示,血铬水平高是CDK6基因过量表达的危险因素(OR=1.168,95%CI:1.040~1.312)。结论六价铬职业暴露可致细胞周期相关基因CDK6表达升高。
Objective To study the effect of hexavalent chromium exposure on the expression of cell cycle related genes in professional population. Methods A total of 155 hexavalent chromium occupational exposure workers in a electroplating factory were selected as the research object to investigate the basic situation of workers and to collect heparin anticoagulant peripheral blood. The level of blood chromium (ppb) was measured by inductively coupled plasma mass spectrometry. Subjects were divided into 4 groups (group 1≤0.04 ppb, group0≤1 ppb, group0≤5 ppb, group4≤5 ppb), comparing the expression levels of p16 and CDK6 genes in each group, and analyzing the gene Influential factors of expression. Results A total of 155 workers were recruited, including 89 males and 66 females, with an average age of 39.65 ± 8.86 years. The median length of service was 36 months. The median blood chromium level was 0.95 ppb. The median of p16 gene expression in four groups was 4.22 (19.91), 7.19 (30.71), 7.47 (21.47) and 14.60 (27.28), respectively. There was no significant difference between the two groups (P = 0.19). The median CDK6 gene expression level in four groups was 4.00 (8.13), 15.05 (32.72), 8.03 (17.32) and 24.81 (35.09), respectively. The CDK6 expression level in group 1 was lower than that in other groups (P <0.01). Spearman correlation analysis showed that there was a positive correlation between CDK6 gene expression and blood chromium levels (rs = 0.28, P <0.01). Logistic regression analysis showed that high blood chromium level was a risk factor for overexpression of CDK6 (OR = 1.168,95 % CI: 1.040 ~ 1.312). Conclusion Occupational exposure to hexavalent chromium leads to an increase in the expression of CDK6, a cell cycle related gene.
