产后抑郁大鼠模型的构建及参归仁合剂对其抑郁样行为的影响

目的:建立产后抑郁症(PPD)大鼠模型,观察参归仁合剂对其抑郁样行为的影响。方法:150只雌性大鼠随机分为空白组、PPD模型组、舍曲林组、雌二醇组、参归仁组(高、中、低剂量),每组设1、2、4周观察时点,分别对其进行行为学测试并观察其抑郁样行为。结果:与空白组比较,PPD模型组各时点大鼠体重增加缓慢,旷场实验水平运动得分、垂直运动得分降低,强迫游泳不动时间显著延长,糖水消耗量明显下降(P<0.001);与PPD模型组比较,参归仁高、中、低剂量组和阳性对照组各项指标均有明显改善(P<0.001或P<0.01)。结论:产后抑郁大鼠表现出抑郁样行为,参归仁合剂能够改善其行为学指标,具有抗抑郁作用。 Objective: To establish a rat model of postpartum depression (PPD) and observe its effect on depression-like behavior. Methods: 150 female rats were randomly divided into blank group, PPD model group, sertraline group, estradiol group and ginseng group (high, medium and low doses) At the same time, behavioral tests and depression-like behaviors were observed respectively. Results: Compared with the blank group, the body weight of rats in PPD model group increased slowly at each time point. The scores of open field exercise, vertical motion, forced swimming were significantly prolonged, and the consumption of carbohydrate decreased significantly (P <0.001). Compared with the PPD model group, the parameters of high, medium and low dose group and positive control group were significantly improved (P <0.001 or P <0.01). Conclusion: Postpartum depression in rats showed depression-like behavior, Shen Gui Ren Mixture can improve their behavioral indicators, with anti-depressive effect.
