目的:研究荆防散乙酸乙酯部位对大鼠、豚鼠过敏性模型及IFN-γ、IL-4水平的影响,初步探讨其抗过敏作用机制与纠正Th1/Th2免疫失衡的关系。方法:采用卵白蛋白合并百白破疫苗致大鼠全身主动过敏反应模型和卵白蛋白致豚鼠哮喘模型,测定大鼠脏器(胸腺、脾)指数及血清IL-4、IFN-γ水平;测定豚鼠全血白细胞计数、血清Ig E水平,及肺泡灌洗液(BALF)中IL-4、IFN-γ水平和总蛋白含量。结果:与模型对照组比较,荆防散乙酸乙酯部位5.0 g生药/kg、2.5 g生药/kg显著降低全身主动过敏反应模型大鼠血清IL-4水平、升高大鼠胸腺指数及血清IFN-γ水平,并能显著降低哮喘模型豚鼠全血白细胞计数和血清Ig E水平;荆防散乙酸乙酯部位2.5 g生药/kg显著对抗哮喘模型豚鼠肺泡灌洗液中IL-4、总蛋白含量的升高及IFN-γ水平的降低。结论:荆防散乙酸乙酯部位抗过敏作用良好,作用机制与上调IFN-γ水平、下调IL-4水平有关,提示其能通过纠正Th1/Th2免疫失衡来抑制过敏反应。
Objective: To study the effects of Jing Fang San Ethyl Acetate on the anaphylactic model and the levels of IFN-γ and IL-4 in rats and guinea pigs, and to explore the relationship between its anti-allergic mechanism and the correction of Th1 / Th2 immune imbalance. Methods: The model of active anaphylaxis in rats and ovalbumin-induced guinea pig asthma model were induced by ovalbumin combined with diphtheria-baek vaccine. The levels of viscera (thymus and spleen), serum IL-4 and IFN-γ in rats were measured. Whole blood leukocyte count, serum Ig E level, and IL-4, IFN-γ levels and total protein content in BALF. Results: Compared with the model control group, 5.0 g crude drug / kg and 2.5 g crude drug / kg at the ethyl acetate fraction of Jing Fang San San significantly decreased the level of serum IL-4 in the systemic active anaphylaxis model rats, increased thymus index and serum IFN- γ level, and can significantly reduce the asthma model guinea pig white blood cell count and serum Ig E levels; Jing antimicrobial ethyl acetate 2.5 g crude drug / kg significantly inhibited the asthmatic model of guinea pig BALF IL-4, total protein content Increase and decrease of IFN-γ level. CONCLUSION: Jing anti-allergic effect of Ethyl acetate fraction is good and its mechanism is related to the up-regulation of IFN-γ and down-regulation of IL-4, suggesting that it can inhibit allergic reaction by correcting Th1 / Th2 immune imbalance.
