目的探讨肿瘤转移抑制基因1(metastasis suppressor 1,MTSS1)及Gli1基因在子宫内膜腺癌细胞系Ishikawa和HEC-1A中的m RNA的转录及蛋白表达水平。方法体外培养子宫内膜高分化腺癌细胞系Ishikawa和中分化腺癌细胞系HEC-1A细胞,采用RT-PCR和Western blot法分别检测两株癌细胞系中MTSS1和Gli1基因m RNA的转录及蛋白的表达水平。结果子宫内膜高分化腺癌细胞系Ishikawa中MTSS1基因m RNA的转录及蛋白的表达水平显著高于中分化细胞系HEC-1A(P<0.05),Ishikawa细胞系中Gli1基因m RNA转录及蛋白表达水平显著低于HEC-1A细胞系(P<0.05)。结论 MTSS1可能作为转移抑制基因,通过调控Sonic hedgehog(Shh)信号通路参与子宫内膜腺癌的发生发展,为进一步研究子宫内膜腺癌的发生发展机制奠定了基础。
Objective To investigate the transcription and protein expression of m RNA of metastasis suppressor 1 (MTSS1) and Gli1 in endometrial adenocarcinoma cell lines Ishikawa and HEC-1A. Methods Ishikawa, a well-differentiated adenocarcinoma cell line, and a moderately-differentiated adenocarcinoma cell line HEC-1A were cultured in vitro. The transcription of m RNA of MTSS1 and Gli1 in two cancer cell lines was detected by RT-PCR and Western blot respectively. Protein expression levels. Results The mRNA and protein expression of MTSS1 mRNA in Ishikawa cell line was significantly higher than that in the moderately differentiated cell line HEC-1A (P <0.05). In the Ishikawa cell line, the m RNA transcription of Gli1 gene and protein The expression level was significantly lower than that of HEC-1A cell line (P <0.05). Conclusions MTSS1 may play a role as a metastasis suppressor gene in the pathogenesis of endometrial adenocarcinoma by regulating the Sonic hedgehog (Shh) signaling pathway, which may lay the foundation for the further study on the mechanism of endometrial adenocarcinoma.
