目的研究石蒜属13种(含2变种)植物的种间亲缘关系和同种不同采集地材料的遗传多样性,为种质资源的开发利用提供分子佐证。方法采用扩增长度多态性(AFLP)标记对石蒜属植物20份材料的基因组DNA进行种间亲缘关系研究。结果从33对选择性扩增引物组合中筛选出6对,共扩增得到330条清晰可统计的条带,其中多态性条带为320条,占总带数的96.8%;AFLP分子标记检测到石蒜属20份材料种间遗传相似系数为0.38~0.89;AFLP分子标记聚类结果显示同一采集地的材料亲缘关系更近;石蒜属植物存在较丰富的遗传多样性。结论 AFLP分子标记能有效区分石蒜属植物种间的亲缘关系。
Aim To study the genetic relationship among 13 species (including 2 varieties) of Lycoris and the genetic diversity of the materials collected from the same species, and to provide molecular evidence for the exploitation and utilization of germplasm resources. Methods The genomic DNA of 20 genus Lycoris from different genus Lycoris was studied by AFLP. Results Sixteen pairs of primers were screened from 33 pairs of primers. A total of 330 clear and statistically detectable bands were amplified, of which 320 were polymorphic bands, accounting for 96.8% of the total bands. AFLP markers The genetic similarity coefficients of the genus Lycoris from 20 species were detected between 0.38 and 0.89. AFLP molecular marker clustering results showed that the materials of the same locus were more closely related. The genus Lycoris had rich genetic diversity. Conclusion AFLP molecular markers can effectively distinguish the genetic relationship among species of Lycoris.
