目的:通过体外实验探讨紫杉醇对恶性胶质瘤9L细胞株有否凋亡诱导作用。方法:在经MTT比色法测试到紫杉醇对9L细胞具有细胞毒作用的前提下,进一步通过形态学观察,DNA琼脂糖凝胶电泳及流式细胞仪来检测其细胞毒作用是否与诱导细胞凋亡有关。结果:紫杉醇对9L细胞具显著细胞毒作用;01μmol/L紫杉醇作用下9L细胞分裂主要被阻滞在G2/M期,并在第72h出现凋亡细胞峰,此时培养细胞HE染色表现出典型的凋亡细胞形态特征,抽提其DNA做琼脂糖凝胶电泳显示凋亡特有的DNA“阶梯”。结论:紫杉醇具有抗胶质瘤作用,其机制与诱导细胞凋亡有关,通过进一步的体内实验,可为其最终广泛应用于临床脑胶质瘤病人提供充分的理论依据。
OBJECTIVE: To investigate whether paclitaxel induces apoptosis in 9L cell line of malignant glioma in vitro. Methods: MTT assay was used to determine the cytotoxic effect of paclitaxel on 9L cells. Morphological observation, DNA agarose gel electrophoresis and flow cytometry were used to detect the cytotoxicity of paclitaxel on the 9L cells. Death related. Results: Paclitaxel had a significant cytotoxic effect on 9L cells. 9L cells were arrested in G2 / M phase under 01μmol / L paclitaxel, and apoptotic cells peak appeared at 72h. HE staining of cultured cells A typical morphological characteristics of apoptotic cells, extracted DNA agarose gel electrophoresis showed apoptosis-specific DNA “ladder.” Conclusion: Paclitaxel has the anti-glioma effect. The mechanism of paclitaxel is related to the induction of apoptosis. Through further in vivo experiments, paclitaxel may provide a sufficient theoretical basis for the clinical application of paclitaxel in clinical glioma patients.
