卡介苗生产中最重要的质检指标—活菌含量测定 ,传统固体培养计数方法时间需 2 1天以上 ,且结果有时不稳定、操作复杂。使用XTT[2 ,3 bis (2 methoxy 4 nitro 5 sulphenyl) (2H) tetrazolium 5 carboxanilide]做底物的四唑钅翁盐试验在活卡介菌的还原下快速显色 ,很好地反映出卡介苗活菌的浓度差异。实验观察在微孔培养板中反应经酶标仪测定的XTT试验非常简便易行 ,不受死亡细菌的影响 ,不受培养基的影响 ,实验重复性非常好 ,检测活性单位时间惊人地缩短为 2 4小时左右 ,检测活性范围大。此方法有潜力代替传统活菌计数法用于卡介苗生产中的活性单位测定。
