目的:研究陇中损伤散对兔激素性股骨头坏死模型的治疗效果及其作用机理。方法:成年青紫蓝兔75只,随机分为6组:正常组、模型组、健骨生丸组、陇中损伤散高、中、低剂量组。采用内毒素加激素诱导的方法建立早期激素性股骨头坏死模型。造模后2周,各组动物给予相应的药物或生理盐水灌胃。用药后2、4、8周心脏采血。第8周末处死动物,取双侧股骨头进行病理组织观察,血浆ET与NO、血浆TXB2与6-keto-PGF1α含量的变化。结果:病理学观察显示:与N组比较,M组骨组织形态学出现典型的坏死表现;各治疗组坏死程度较M组显著降低,特别是SH组尤为明显。与M组比较,第8周SH组和SM组ET水平明显降低(P(0.01),而NO水平明显增高(P(0.05)。与M组比较,第2到8周SH组和SM组血浆TXB2含量明显下降,而6-keto-PGF1α含量明显上升,且第8周两者都基本接近正常值水平(P(0.05或P(0.01)。SH组和SM组的T/K比值较M组有显著减小,用药后第2到8周,相应各组指标M组与N组比较差异显著(P(0.05或P(0.01),Y组与M组比较也有显著性差异(P(0.05或P(0.01)。结论:陇中损伤散具有治疗早期激素性股骨头坏死的作用,其机理可能是部分改善血管内皮功能受损状态,降低血液黏稠度,降低骨内压,改善微循环。
OBJECTIVE: To study the therapeutic effect and mechanism of Suizhong Powder on rabbit model of femoral head necrosis. METHODS: Seventy-five adult cyanotic blue rabbits were randomly divided into 6 groups: normal group, model group, Jiangu Shengwan group, middle lozenge group, middle and low dose group. Early hormone-induced femoral head necrosis models were established using endotoxin plus hormone-induced methods. Two weeks after modeling, each group of animals was given intragastric administration of the corresponding drug or saline. Blood was collected from the heart at 2, 4 and 8 weeks after administration. At the end of the 8th week, the animals were sacrificed and bilateral femoral heads were taken for histopathological examination. Changes in plasma ET and NO, plasma TXB2 and 6-keto-PGF1α levels were observed. RESULTS: Pathological observations showed that compared with group N, typical histological features of necrosis were observed in group M. The degree of necrosis in each treatment group was significantly lower than that in group M, especially in group SH. Compared with group M, ET levels in SH group and SM group were significantly decreased at week 8 (P(0.01), and NO level was significantly higher (P(0.05). Compared with group M, plasma from SH group and SM group from week 2 to 8) The content of TXB2 decreased significantly, while the content of 6-keto-PGF1α increased significantly. At the 8th week, both levels were almost close to the normal level (P(0.05 or P(0.01)). The T/K ratio of SH group and SM group was higher than that of M group. Significantly decreased, from the 2nd to the 8th week after administration, the corresponding indicators in each group were significantly different from those in the N group (P(0.05 or P(0.01)). There was also a significant difference between the Y group and the M group (P(0.05 or P (0.01).CONCLUSION: Suizhong Decoction has the effect of early steroid-induced femoral head necrosis, and its mechanism may be partly to improve the state of vascular endothelial dysfunction, reduce blood viscosity, reduce intra-bone pressure, and improve microcirculation.
