目的探讨高尔基体α-甘露糖苷酶Ⅱ(GMⅡ)和E-钙黏素(E-cadherin)在正常肝组织及不同分化的肝癌组织中的表达及其意义。方法采用RT-PCR法检测GMⅡ和E-cadherin mRNA在正常肝组织、高分化及中低分化肝癌组织中的表达;免疫组化SP法及Western blot法检测GMⅡ和E-cadherin蛋白在3种肝组织中的表达,并对GMⅡ和E-cadherin的差异表达进行相关性分析。结果 RT-PCR及Western blot分析显示,GMⅡmRNA和蛋白在正常肝组织中表达水平最低,在中低分化肝癌组织中表达水平最高,在高分化肝癌组织中表达居中,E-cadherin的表达则相反,各组间表达差异均有统计学意义(P<0.05);免疫组化SP法检测显示,GMⅡ主要表达于正常肝组织及肝癌组织的胞浆,在正常肝组织、高分化及中低分化肝癌组织的表达阳性率分别为40%、64%和86%,E-cadherin在正常肝组织的胞膜、胞浆中均有表达,而在肝癌组织中则主要表达于胞浆,其表达阳性率分别为70%、33%和9%,各组间表达差异均有统计学意义(P<0.05);GMⅡ和E-cadherin表达的Pearson相关系数r值为-0.415,P值为0.01。结论 GMⅡ和E-cadherin在肝癌组织中的表达呈负相关,GMⅡ对肝癌的发生发展可能发挥一定的作用。
Objective To investigate the expression and significance of golgi α-mannosidase Ⅱ (GMⅡ) and E-cadherin in normal liver tissue and differentiated hepatocellular carcinoma. Methods The expressions of GMⅡ and E-cadherin mRNA in normal liver tissue, well differentiated and moderately differentiated HCC tissues were detected by RT-PCR. The expressions of GMⅡ and E-cadherin protein were detected by immunohistochemical SP method and Western blot in 3 kinds of liver The expression of GMⅡ and E-cadherin in different tissues was analyzed by the correlation analysis. Results RT-PCR and Western blot showed that the expression of GMⅡmRNA and protein was the lowest in normal liver tissues, highest in poorly differentiated HCC tissues, moderately expressed in well-differentiated HCC tissues, and negative in E-cadherin, There were significant differences among the groups (P <0.05). The immunohistochemical SP method showed that the expression of GMⅡ was mainly in the cytoplasm of normal liver tissues and hepatocellular carcinoma tissues. In normal liver tissues, well differentiated and moderately poorly differentiated hepatocellular carcinoma The expression rates of E-cadherin in normal liver tissues were 40%, 64% and 86% respectively.
