本文利用Sephacry1S 2 0 0和Q Sepharose两步层析 ,从HP蒸馏水浸液中提取纯化尿素酶 ,并对其特性进行了测定 ,证明HP尿毒酶各含一个 6 6kD和 2 9 5kD的亚单位 ,并以 6个分子聚合成 6 2 5kD大分子蛋白 ,有很好的抗原性 ,并显示特异的血清学反应。用HP超声浸液抗原和尿素酶 ,加入 2 μg霍乱毒素粘膜佐剂 ,给SPFBALB/C小鼠口服免疫 ,可使 80 %小鼠抵抗HP活菌的攻击 ,证明尿素酶是一种抗HP的保护性抗原。
In this paper, purified urease was extracted from aqueous extract of HP by Sephacryls 2000 and Q Sepharose two-step chromatography, and its characteristics were determined. It was proved that each of the urease contained 6 subunits of 6 6 kD and 2 9 5 kD, And 6 molecules aggregated into 625kD macromolecular protein, has good antigenicity, and showed specific serological response. HP hypersonic infusion of antigens and urease, adding 2 μg of cholera toxin mucosal adjuvant, oral immunization of SPFBALB / C mice, 80% of mice can be resistant to HP live bacteria attack, proved that urease is an anti-HP Protective antigen.
