目的研究透明质酸(HA)对鼻咽癌细胞体内外增殖特性的影响。方法以鼻咽癌CNE-2Z细胞为对象,采用增殖实验、裸鼠体内成瘤实验、图像分析及免疫组化等方法,观察HA对癌细胞体内外增殖力的影响。结果增殖实验表明HA在浓度为0.5mg、1mg、2mg范围内,增殖A比值分别为1.89±0.23、2.09±0.26、2.58±0.59,与对照组A值比较有高度显著性差异(P<0.01),且呈量效正相关。HA可使鼻咽癌细胞裸小鼠移植瘤成瘤早,生长快,实验组移植瘤平均潜伏期、倍增时间较对照组显著缩短(P<0.01);裸鼠移植瘤组织切片PCNA免疫组化染色结果显示,实验组细胞PCNA阳性率、阳性面积明显高于对照组,两组比较有显著性差异(P<0.01);平均光密度高于对照组,有显著性差异(P<0.05)。结论HA对鼻咽癌细胞体外增殖及裸鼠移植瘤的生长有明显促进作用,有助于进一步研究鼻咽癌的侵袭和转移机理
Objective To investigate the effect of hyaluronic acid (HA) on proliferation of nasopharyngeal carcinoma cells in vitro and in vivo. Methods Nasopharyngeal carcinoma CNE-2Z cells were treated with proliferation assay, tumorigenesis in nude mice, image analysis and immunohistochemistry to observe the effect of HA on proliferation of cancer cells in vitro and in vivo. Results Proliferation experiments showed that the proliferation A ratio of HA at concentration of 0.5mg, 1mg and 2mg was 1.89 ± 0.23, 2.09 ± 0.26 and 2.58 ± 0.59, respectively. Compared with the control group A value was highly significant difference (P <0.01), and the amount of positive correlation. HA could make nasopharyngeal carcinoma xenografts in nude mice grow early and grow fast, the average incubation period and doubling time of experimental xenografts were significantly shorter than those in control group (P <0.01); The expression of PCNA in nude mice The results of staining showed that the positive rate of PCNA in experimental group was significantly higher than that in control group (P <0.01), and the average optical density was higher than that in control group (P < 0.05). Conclusion HA can promote the proliferation of nasopharyngeal carcinoma cells in vitro and the growth of xenografts in nude mice, which is helpful to further study the mechanism of invasion and metastasis of nasopharyngeal carcinoma
