采用左心室及血管插管法,研究了洛土辛(Lot)对大鼠及狗血流动力学的影响。 Lot 2.5,5,10 mg/kg,iv可剂量依赖性地增加麻醉大鼠的左心室压(LVP)、左室压力变化最大速率(±dp/dtmax)、动脉收缩压(SAP),降低左室舒张末期压(LVEDP),但对心率(HR)和动脉舒张压(DAP)却无明显的影响。麻醉大鼠 Lot 5 mg/kg、麻醉狗 Lot 3 mg/kg,iv可获相似结果,其对狗血流动力学影响的起效时间较快且作用持续较长。在毁脊髓大鼠,Lot 5 mg/kg,iv不仅显著地增加 LVP,±ap/dtmax,SAP,明显增加DAP和HR。提示Lot对血流动力学影响的特性与PDE Ⅲ抑制剂相似。
Left ventricle and vessel intubation were used to study the effect of Lotus on hemodynamics in rats and dogs. Lot 2.5,5,10 mg/kg,iv dose-dependently increased left ventricular pressure (LVP), maximum rate of left ventricular pressure change (±dp/dtmax), and arterial systolic pressure (SAP) in anesthetized rats. Left ventricular end-diastolic pressure (LVEDP) was reduced, but there was no significant effect on heart rate (HR) and arterial diastolic pressure (DAP). Anesthetized rat Lot 5 mg/kg, anesthetized dog Lot 3 mg/kg,iv can obtain similar results, and its onset time for dog hemodynamics is faster and lasts longer. In spinal cord-destroying rats, Lot 5 mg/kg, iv not only increased LVP, ±ap/dtmax, SAP, but also significantly increased DAP and HR. It is suggested that the effect of Lot on hemodynamics is similar to that of PDE III inhibitors.
