目的:观察哮喘豚鼠经β-细辛醚喷雾给药后,对白细胞介素-4(IL-4)、白细胞介素-16(IL-16)和γ-干扰素(γ-IFN)及小鼠免疫器官指数的影响。方法:将豚鼠分为高、中、低剂量组,阳性药组和模型组,复制哮喘模型,造模7d后,喷雾给药,7d后引喘,记录哮喘发作潜伏期和跌倒潜伏期。取血测定IL-4、IL-16和γ-IFN水平。另取小鼠分为高、中、低剂量组,阳性药组和正常组,喷雾给药7d后,摘取胸腺、脾脏,计算免疫器官指数。结果:β-细辛醚能延长哮喘发作潜伏期和跌倒潜伏期(P<0.01);能降低IL-4的含量、提高IL-16和γ-IFN水平,降低IL-4/γ-IFN比值(P<0.05,P<0.01);能提高小鼠胸腺指数和脾脏指数(P<0.05,P<0.01)。结论:β-细辛醚对哮喘有整体治疗作用;有一定的免疫兴奋作用,并有一定的剂量依赖性,其治疗哮喘可能是通过调节Th1/Th2平衡而实现的。
OBJECTIVE: To observe the effects of β-asarone on the expression of interleukin-4 (IL-4), interleukin-16 (IL-16) and γ-interferon Effect of mouse immune organ index. Methods: Guinea pigs were divided into high, medium and low dose groups, positive drug group and model group. The model of asthma was replicated. After 7 days of modeling, guinea pigs were sprayed for administration and asthma was induced after 7 days. The incubation period and fall latency of asthma were recorded. Blood samples were taken for determination of IL-4, IL-16 and γ-IFN levels. The other mice were divided into high, medium and low dose groups, positive drug group and normal group. After spraying for 7 days, the thymus and spleen were harvested and the immune organ index was calculated. Results: β-asarone prolonged the latent period and fall latency of asthma (P <0.01), decreased the content of IL-4, increased the levels of IL-16 and γ-IFN and decreased the ratio of IL-4 / <0.05, P <0.01); can improve the thymus index and spleen index of mice (P <0.05, P <0.01). CONCLUSION: β-asarone has an overall therapeutic effect on asthma; it has some immunostimulatory effects and a certain dose-dependent manner. Its treatment of asthma may be achieved through the regulation of Th1 / Th2 balance.
