目的:检测T淋巴瘤TCR β链独特型DNA疫苗的基因表达。方法:大剂量提取质粒,将BALB/c鼠随机分为 pcDNA3.l组、pcDNA3.1/TCR Vβ8组、pcDNA3.1/TCR Vβ8+IL-2组、pcDNA3.l/TCR Vβ8+CpG+脂质体组和全硫代CpG组共5组(每组6只),双侧股四头肌内注射疫苗免疫,于第0、2、4周各免疫1次,共3次。接种疫苗后 5 d,用 RT-PCR法检测重组质粒mRNA表达。接种疫苗后 7 d应用免疫组化检测重组质粒蛋白表达。结果:pcDNA3.1/TCR Vβ8组、pcDNA3.l/TCR Vβ8+IL-2组、pcDNA3.l/TCR Vβ8+CpG+脂质体组小鼠骨骼肌中均显示mRNA和蛋白表达,而pcDNA3.1组和全硫代CpG组未见mRNA和蛋白表达。结论:T淋巴瘤TCRβ链独特型DNA疫苗在小鼠骨骼肌中有效地表达了基因产物。
Objective: To detect the gene expression of TCR β chain idiotypic DNA vaccine in T lymphoma. Methods: The plasmid was extracted by high dose and BALB / c mice were randomly divided into pcDNA3. l group, pcDNA3.1 / TCR Vβ8 group, pcDNA3.1 / TCR Vβ8 + IL-2 group and pcDNA3. 5 mice in each group (6 mice in each group) received intraperitoneal injection of quadriceps intramuscular immunization and immunized once weekly for 0, 2 and 4 weeks. A total of 3 times. Five days after vaccination, recombinant plasmid mRNA expression was detected by RT-PCR. Seven days after vaccination, recombinant plasmids were detected by immunohistochemistry. Results: pcDNA3.1 / TCR Vβ8 group, pcDNA3. l / TCR Vβ8 + IL-2 group, pcDNA3. l / TCR Vβ8 + CpG + liposome group mouse skeletal muscle both mRNA and protein expression, and pcDNA3.1 group and the whole thio CpG group no mRNA and protein expression. CONCLUSION: The TCR β-chain idiotypic DNA vaccine of T lymphoma effectively expresses the gene product in mouse skeletal muscle.
