目的 研究慢性胆囊疾患胆囊上皮细胞K ras、P5 3基因变异情况 ,进行癌前病变的预测和监测。方法 取手术切除慢性胆囊疾患的胆囊 ,福尔马林固定、石蜡包埋连续切片 ,HE染色 ,显微镜下微解剖选取癌前病变部位 ,立体显微镜下回收组织细胞 ,抽提DNA行K ras、P5 3的PCR SSCP分析。结果 检测 11例 ,9例K ras基因有变异 ,其中ex on 1变异 7例 ,exon 1和exon 2变异 2例 ;P5 3基因的exon 5~ 9未检测到变异。结论 采用显微解剖并PCR SSCP分析可进行胆囊癌前病变基因变异研究 ,实现癌前监测 ,适时确立手术时机 ,对胆囊癌的防治具有重要意义
Objective To study the variation of K ras and P53 gene in gallbladder epithelial cells in patients with chronic gallbladder disease and to predict and monitor precancerous lesions. Methods The gallbladder, formalin-fixed and paraffin-embedded serial sections of chronic gallbladder disease were removed by surgery. The sections were stained with HE, and the precancerous lesion was selected by microsurgical dissection. Tissue cells were harvested under stereomicroscope. 3 PCR SSCP analysis. Results There were 11 cases and 9 cases of K ras gene variation. There were 7 cases of ex on 1 mutation, 2 cases of exon 1 and exon 2 mutation, and 5 cases of exon 5 -9 of P5 gene gene. Conclusion Microscopic anatomy and PCR SSCP analysis of gallbladder precancerous lesions genetic variation study, to achieve precancerous monitoring, timely establishment of operation timing, the prevention and treatment of gallbladder cancer is of great significance
