目的 :研究切应力对与血管平滑肌细胞联合培养的内皮细胞的抗凝血和粘附的影响。方法 :对与平滑肌细胞联合培养的内皮细胞施加生理范围的切应力 ,用免疫组化及图像分析法检测内皮细胞的血管假性血友病因子 (v WF)含量。 结果 :联合培养的内皮细胞 v WF的变化与切应力大小及作用时间有关 ,随切应力增大 ,胞内 v WF含量逐渐减少 ,且切应力作用的第 1小时减少的最快。结论 :生理范围的切应力作用 ,可能减少与平滑肌细胞联合培养的内皮细胞的 v WF表达 ,或促使 v WF短时释放加快 ,有利于增强抗凝血功能及内皮细胞的粘附。
Objective: To study the effect of shear stress on the anticoagulant and adhesion of endothelial cells cultured with vascular smooth muscle cells. METHODS: Physiological shear stress was applied to endothelial cells cultured in combination with smooth muscle cells. The content of von Willebrand factor (v WF) in endothelial cells was detected by immunohistochemistry and image analysis. Results: The change of vWF in cultured endothelial cells was related to the magnitude of shear stress and the duration of action. With the increase of shear stress, the content of intracellular vWF decreased gradually, and the shear stress decreased rapidly at the first hour. CONCLUSIONS: Physiological shear stress may reduce the vWF expression in endothelial cells co-cultured with smooth muscle cells or accelerate the short-term release of vWF, which may be beneficial for enhancing anticoagulant function and endothelial cell adhesion.
