目的:探讨蚝菇提取物对老年大鼠(24月龄)肝肾组织内过氧化物酶基因表达及蛋白质羰基含量的影响。方法:18只大鼠被分为3组,每组6只。组1为健康年轻大鼠(4月龄)对照组;组2为健康老年大鼠(24月龄)对照组;组3为健康老年大鼠(24月龄)蚝菇提取物干预组,干预措施为每天腹腔注射蚝菇提取物200 mg/kg体质量,连续注射30 d。第31天处死全部大鼠并取出肝肾组织。使用逆转录聚合酶链反应及分光光度测定法分别测定大鼠肝肾组织内过氧化物酶基因表达情况及蛋白质羰基的含量。结果:组3老年大鼠注射蚝菇提取物后,其肝肾组织内过氧化物酶基因表达显著高于组2老年大鼠,且组3老年大鼠肝肾组织内蛋白羰基含量显著低于组2老年大鼠。结论:蚝菇提取物能够增加老年大鼠肝肾组织内抗氧化酶基因的表达并减少自由基引起的蛋白质氧化,对于自由基引起的年龄相关性疾病可能有保护作用。
Objective: To investigate the effects of oyster mushroom extract on peroxidase gene expression and protein carbonyl content in liver and kidney of aged rats (24 months old). Methods: Eighteen rats were divided into 3 groups with 6 rats in each group. Group 1 was a control group of healthy young rats (4 months old); Group 2 was a control group of healthy aged rats (24 months old); Group 3 was an oyster mushroom extract intervention group of healthy old rats (24 months old) Measures were daily intraperitoneal injection of oyster mushroom extract 200 mg / kg body weight, continuous injection 30 d. On the 31st day, all the rats were sacrificed and liver and kidney tissues were removed. Reverse transcriptase polymerase chain reaction and spectrophotometry were used to determine the expression of peroxidase gene and the content of protein carbonyls in rat liver and kidney. Results: The expression of peroxidase gene in liver and kidney tissues of group 3 senile rats injected with oyster mushrooms was significantly higher than that of group 2 senile rats, and the protein carbonyl content in liver and kidney of group 3 senile rats was significantly lower than that of group 2 Group 2 aged rats. Conclusion: Oyster mushroom extract can increase the antioxidant enzyme gene expression in liver and kidney tissues of aged rats and reduce the protein-induced oxidative stress induced by free radicals, which may have protective effects on the free radical induced age-related diseases.
