目的:探讨不同肝组织中VEGF-B及其两个变位剪切体VEGF-B167、VEGF-B186mRNA的表达,并评估其临床意义.方法:应用荧光定量PCR法检测肝癌、癌旁、乙型肝炎肝硬化、慢性乙型肝炎及正常肝组织中总VEGF-B、VEGFB167及VEGFB186mRNA的表达量,与患者的临床病理资料做相关性分析,并结合随访资料做生存分析.结果:总VEGF-B及VEGF-B186mRNA在各组组织中表达量无统计学差异;VEGF-B167mRNA在各组间表达量有统计学差异,正常组表达最高,乙型肝炎肝硬化组最低(252.59vs168.00,P=0.005),肝癌组中VEGF-B167的表达量明显高于慢性乙型肝炎组和乙型肝炎肝硬化组(234.13vs170.72,168.00;P=0.043,P<0.01).依据VEGF-B表达高低,将肝癌患者分组,VEGF-B167低表达组患者的总体生存率较高表达组和无差异组有升高趋势(P=0.097),而VEGF-B167低表达组患者术后无瘤生存率明显高于其他两组(P=0.023).肝癌组织中总VEGF-B、VEGF-B167及VEGF-B186的表达与患者性别、年龄、病毒感染、肝硬化、Child-Pugh分级、AFP值、肿瘤数目、肿瘤大小、血管侵犯、TNM分期及肿瘤分化之间相关性无统计学意义.结论:VEGF-B参与了肝癌的发生发展过程,VEGF-B167在此过程中的作用较VEGF-B186更重要.VEGF-B167低表达患者预后较好,术后总生存率和无瘤生存率较高.
Objective: To investigate the expression of VEGF-B16 and VEGF-B186 mRNA in different liver tissues and evaluate the clinical significance of VEGF-B167 and VEGF-B186.Methods: The expression of VEGF-B167 and VEGF- The expression of VEGF-B, VEGFB167 and VEGFB186 mRNA in cirrhosis, hepatitis B cirrhosis, chronic hepatitis B and normal liver tissues were analyzed with the clinicopathological data of the patients and the follow-up data were used for survival analysis.Results: The total VEGF-B And VEGF-B186mRNA expression in each group had no statistical difference; VEGF-B167mRNA expression between the groups were statistically different, the highest expression in normal group, hepatitis B cirrhosis group the lowest (252.59vs168.00, P = 0.005).
