目的研究E2F1、MDM2在不同级别恶性人脑胶质瘤中表达的水平及其与肿瘤恶性程度的关系。方法共82例胶质细胞瘤标本(I级胶质瘤20例、Ⅱ级胶质瘤20例、Ⅲ级胶质瘤21 例、Ⅳ级胶质瘤21例)进行E2F1、MDM2免疫组化染色。实验结果用统计软件SPSS11.0进行卡方检验。结果 E2F1在低级别(I级、Ⅱ级)胶质瘤中的阳性表达率较高,在高级别(Ⅲ级、Ⅳ级)胶质瘤中较低,两者相比差别有显著性意义(X2=3.919,P<0.05);而MDM2在低级别胶质瘤中阳性表达率较低, 在高级别胶质瘤中较高,两者相比差别有极显著性意义(X2=12.602,P<0.01)。结论 E2F1和MDM2 在胶质瘤中所起的作用不同,前者在胶质瘤的发生发展中可能起了抑癌基因的作用,而后者在其中扮演了癌基因的角色。
Objective To investigate the expression of E2F1 and MDM2 in malignant human gliomas of different grades and their relationship with malignancy of tumor. Methods A total of 82 glioma specimens (20 grade I gliomas, 20 grade II gliomas, 21 grade III gliomas and 21 grade IV gliomas) were immunohistochemically stained for E2F1 and MDM2 . Experimental results using statistical software SPSS11.0 chi-square test. Results The positive expression rate of E2F1 in gliomas of low grade (Grade I, Grade II) was higher than that in grade Gliomas (Grade III and IV), with significant difference (P < X2 = 3.919, P <0.05). However, the positive expression rate of MDM2 in low-grade glioma was lower and higher in high-grade gliomas, with significant difference (X2 = 12.602, P <0.01). Conclusion The role of E2F1 and MDM2 in glioma is different. The former may play a role of tumor suppressor gene in the development of glioma, while the latter plays an oncogene role.
